How to read this
The red mark is the published kill line — the price that would prove the thesis wrong. The dot is where the name closed on 22 October 2026; a dot left of the mark has closed through its line.
Distance is drawn on a square-root scale, so close calls get the room. Past 8% a row reads well clear, with a hollow dot to show the figure is off the drawn scale. Rows run tightest first.
Kill line
A daily close below $3.40, a round-number level under the 22 October 2026 close of $3.63. An MGC026 poster without cohort-level confirmed response counts and duration, or one reporting treatment-related lung or eye toxicity, would also defeat the clinical case.
Not a pickResearch coverage at low conviction. A medium-or-higher call would open a pick on the scoreboard.

How picks are graded →The thesis
MacroGenics' cancer-drug candidate MGC026 still needs confirmed, durable head-and-neck tumor responses to justify a revaluation, and the poster scheduled for 23 October 2026 at the European Society for Medical Oncology (ESMO) congress is the event that settles it. The shares closed at $3.63 on 22 October 2026, 25.3% below their $4.86 52-week high.
The clinical claim still rests on one company statement. On 23 July 2026, MacroGenics said the head-and-neck cohort of its Phase 1 study met its prespecified initial response threshold and moved into a second stage of a planned 40-patient cohort. The company has not published confirmed response counts or response duration. Poster 1020P is listed for 15:15 to 16:00 Central European Summer Time on 23 October 2026, and no poster results appeared in company releases or search results as of this note.
One item is new since the prior note of 10 October 2026. On 15 October 2026, MacroGenics announced it had dosed the first patient in a Phase 1 study of MGC030, an antibody-drug conjugate against an undisclosed target, in advanced solid tumors. That met the company's 13 August 2026 expectation of a third-quarter start. It adds a pipeline program without changing the MGC026 efficacy question, so the standing rating is unchanged.
The narrative is maturing: the story is well known, and the shares have drifted from $3.74 on 9 October 2026 to $3.63 on 22 October 2026 into the poster.
Bullish and bearish views on MacroGenics, Inc.
The bull case on MacroGenics, Inc. (MGNX), in brief: The head-and-neck cohort advanced. The bear case: Response evidence is incomplete.
Bull case
- The head-and-neck cohort advanced. MacroGenics reported on 23 July 2026 that the cohort met its prespecified response threshold and entered Stage 2 under a two-stage design. Poster 1020P on 23 October 2026 is the first venue where cohort-level detail can be checked.
- No severe lung or eye toxicity reported so far. As of 8 July 2026, MacroGenics had enrolled 74 patients across dose escalation and expansion. It reported no interstitial lung disease or ocular toxicity, two problems that have affected other drugs of this class.
- A second antibody-drug conjugate has entered the clinic. The 15 October 2026 first-patient dosing of MGC030 met the company's third-quarter timing. The company also expects preliminary MGC028 dose-escalation results in late 2026.
- Funding runs through 2028 on company guidance. MacroGenics reported $173.3 million of cash and securities on 30 June 2026 and $327 million pro forma on 13 August 2026. The pro forma figure includes the $122.5 million sale of manufacturing operations announced in July and expected partner proceeds.
- Sell-side support exists. B. Riley Securities maintained a Buy rating and raised its price target to $11 on 17 August 2026, according to Benzinga. That target is the firm's own view and sits far above the $3.63 close.
Bear case
- Response evidence is incomplete. The 23 July 2026 update contained no confirmed response count or duration figure. A poster that again omits them would leave the revaluation case without its central number.
- Early safety data cover a short window. The 74-patient snapshot dates from 8 July 2026. A treatment-related lung or eye event, or an enrollment halt, in the ESMO update would damage the clinical case.
- Revenue leans on partner payments. Second-quarter 2026 revenue was $32.8 million, with the increase primarily driven by a $24.5 million Sanofi regulatory milestone, per the 13 August 2026 results. Search-aggregated consensus data cited a revenue estimate of $44.04 million, so the quarter came in below it.
- The runway figure is not all cash on hand. The 13 August 2026 guidance through 2028 relies on pro forma cash and projected partner payments. Cash on the balance sheet at 30 June 2026 was $173.3 million.
- The shares have not rallied into the event. The 22 October 2026 close is 9.3% lower than three months earlier, and the 14-day relative strength index (a 0-to-100 momentum gauge) is 51.6. The shares sit well below their 52-week high with no sign of pre-event buying.
Price setup
The adjusted daily close on 22 October 2026 was $3.63. The 52-week high is $4.86, so the shares trade 25.3% below it. The three-month price change is negative 9.3%.
The relative strength index reads 51.6, which is neutral. There is no wide gap above a rising moving average and no sign of crowding in the price. The shares closed at $3.74 on 9 October 2026, and the $3.40 level named below is a round number under the recent closes.
Crowding evidence is limited to the calendar. A scheduled poster on 23 October 2026 makes the event risk imminent, and a quarterly report is likely in November. The research found no SEC filings in the period and no reported insider selling or share issuance.
Catalysts, next 30 days
- ~12 November 2026, est. Third-quarter 2026 results. Date not confirmed by the company; the second-quarter report came on 13 August 2026. It shows cash, partner revenue and any update on the runway guidance.
- Late 2026 — MGC028 preliminary dose-escalation results. Company guidance gives no exact date; this sits outside the 30-day window.
Elapsed catalysts
- 23 October 2026 — MGC026 interim Phase 1 poster 1020P at ESMO. First check of cohort-level confirmed responses, response duration and updated safety behind the head-and-neck threshold claim. (passed 14d ago)
- 26 October 2026 — Lorigerlimab poster 1278P at ESMO. Interim data from the Phase 2 LINNET study in gynecologic cancers add evidence on a separate program without resolving the MGC026 question. (passed 11d ago)
What would change the view
An MGC026 poster that lacks cohort-level confirmed response counts and duration would defeat the clinical case. So would a report of interstitial lung disease, ocular toxicity or a treatment-related enrollment halt. Responses that do not persist would carry the same weight.
The price condition is a daily close below $3.40, a round-number level under the 22 October 2026 close of $3.63. A poster with confirmed, durable head-and-neck responses and no new safety signal would strengthen the case instead. The research has not yet seen poster content, so the rating stays where the evidence leaves it.
What it trades with
MacroGenics trades on its own clinical news rather than on a peer group; no current theme carries the name. Other B7-H3-directed antibody-drug conjugates are the closest read-through, and their data at the same congress would shape sentiment toward MGC026. Small-cap biotech indices set the broader backdrop for financing risk.
Notes
- MacroGenics' 13 August 2026 runway-through-2028 guidance relies on pro forma cash and projected future partner payments, not solely on its 30 June 2026 balance-sheet cash.
- MacroGenics moved to outsourced manufacturing after its July 2026 sale of manufacturing operations for $122.5 million; later development costs are not comparable with earlier periods.
